Restriction digests of the clone give the following sizes (kb): EcoRI--4.6, 3.0; HindIII--4.6, 1.7, 1.3; EcoRI/HindIII--4.6, 1.7, 1.3; BamHI--7.8; PstI--5.2, 2.7. EcoRI detects the cognate Y-specific band (2.8 kb) at high stringency (0.1X SSC, 68C); low stringency detects numerous (10-15) Y-specific bands and one 2 kb autosomal band. TaqI detects 5 independent allelic series representing at least 16 haplotypes, each defined by a different combination of the various forms of each of 5 RFLPs. For TaqI RFLPs, use low stringency (2X SSC, 42C). |
