Restriction digests of the clone give the following sizes (kb): BamHI--19.4, 8.8 doublet, 6.5, 2.7, 0.8; EcoRI--15, 8.8, 7.5, 4.3, 3.4, 2.5, 2.1, 1.9, 1.4, 1.2, 1.1, 1, 0.8; SalI--greater than 25; BglI--11, 7.5, 3.8, 2.1, 1.7, 1.3, 0.8. IMPORTANT: To prevent amplification of a rearranged and/or deleted cosmid, we recommend streaking on LB + amp plates at 30C and picking small colonies for liquid culture. Probe used was a 7.6 kb TaqI fragment. Probe must be prehybridized with excess human DNA to compete out repeated sequences. Loci A, C, and D are in strong linkage disequilibrium. 29/40 (72%) individuals show heterozygosity at 1 or more loci. EcoRI detects an insertion-deletion polymorphism also detected by BamHI and BstEII. |
