Restriction digests of the clone give the following sizes (kb): NotI--16.0, 3.0; SalI--16.0, 3.4; PstI--4.0, 3.9, 2.2, 1.9, 1.85, 1.4, 0.9 + smaller; BglI--3.1, 2.4, 2.3, 2.2, 2.0, 1.4 (doublet), 1.3, 0.9, 0.85 + smaller; BamHI--11.5, 4.6, 2.9. SalI cuts the plasmid in the vector MCS and in intron 1 of the minigene. SstII cuts toward the 3' end of the transferrin promoter and in exon 1 of the minigene. The NotI insert consists of the mouse transferrin promoter (a 3.0 kb BamHI fragment), the human LDL receptor-coding minigene (12.1 kb), and the human growth hormone termination region (0.62 kb). The LDLR minigene consists of separate exons 1, 2, 3, and 4, and fused exons 5-18. |
