Restriction digests of the clone give the following sizes (kb): HindIII--5.9 (doublet), 4.4, 3.5, 0.88; EcoRI--8.2, 6.6, 4.6, 3.1; TaqI--3.6, 2.0, 1.8, 1.5, 1.15 + smaller; BamHI--19.0, 3.2. Codominant segregation demonstrated in 50 3-generation families. IMPORTANT: To prevent amplification of a rearranged and/or deleted cosmid, we recommend streaking on LB + amp plates at 30C and picking small colonies for liquid culture. The probe was pre-associated with excess human DNA before hybridization. Otherwise, RFLPs were observed under normal hybridization and wash stringencies. |
