The EcoRI + BamHI double digest generates a 324 bp fragment by cutting within the polylinkers. Restriction digests of the clone give the following sizes (kb): EcoRI/BamHI--2.9, 0.3; BglI--3.2; BglII--uncut; HindIII--3.2. The antibodies encoded by myeloma MOPC-167 are specific to phosphorylcholine. The insert was cloned from p167kappa RI, and spans the leader--Vk167 region. The T7 promoter is used to make anti-sense RNA. |
