The SnaBI site, in the intron of SUP4ochre, inactivates SUP4ochre.

pYAC3 was constructed from pYAC2 by replacing the SmaI site in the SUP4 intron with the SnaBI site, and eliminating the SfiI and NotI sites flanking the SUP4 gene.
The HIS3 sequence is removed when producing the telomers by BamHI digestion.
