Biosafety Level
1
Host
Distribution host: Escherichia coli HB101
Vector Information
Size (kb): 9.8999996185302730
Vector: pKBY2 (cosmid)
Construction: pBR329, Aspergillus nidulans trpC, pJB8 cos
IMPORTANT: To prevent amplification of a rearranged and/or deleted cosmid, we recommend streaking on LB + amp plates at 30C and picking small colonies for liquid culture.
This is a cosmid vector for isolating genes by complementation of mutations in Aspergillus nidulans. It can also be used for library construction.
A 2.1 kb HindIII/SalI fragment (pJB8) containing lambda cos was ligated to HindIII/SalI-digested pBR329. A 4.1 kb XhoI fragment with wild-type Aspergillus nidulans trpC was ligated into the SalI, and a BamHI linker into the NruI site of pBR329.
The BamHI site was removed from trpC by sodium bisulfite mutagenesis.
Media
ATCC® Medium 1227: LB Medium (ATCC medium 1065) with 50 mcg/ml ampicillin
Growth Conditions
