The insert contains the following restriction sites (with nucleotide positions according to the published sequence): PstI--186 (close to 5' end of the insert), HindIII--246, SstI--403, RsaI--448, 493. Restriction digests of the clone give the following sizes (kb): PstI--4.6; EcoRI--4.6; PvuII--4.6; PstI/EcoRI--3.7, 0.9; PstI/PvuII--3.4, 1.4. The insert cannot be excised by PstI alone due to a deletion in the vector. Approximately 260 bp can be cut out using a PstI/RsaI double digest. Can be used in Northern blots to detect mRNA abundant in chronic myelogenous leukemia but not in normal leukocytes. |
