Restriction digests of the clone give the following sizes (kb): XbaI/BamHI--2.5, 0.75, 0.5; HincII--2.8, 0.85; BglII--3.7, HindIII--3.7, BglII/BamHI--2.2, 1.5.
The XbaI site is upstream of the cDNA insert so excision with this enzyme will include some vector sequence.

Because the host expresses T7 RNA polymerase, and the cDNA is inserted downstream of the phi10 T7 promoter, this strain shows high levels of expression. The product is the mature protein and does not require in vitro cleavage of a fusion protein.
