Restriction digests of the clone give the following sizes (kb): BamHI--9.4, 6.7; EcoRI--6.7 (doublet), 2.3; BglII--12.0, 4.0.
The vector was constructed by inserting the EcoRI fragment from YRp7 containing TRP1 and ARS1 into the SalI site of YEp24.
Constructed by inserting a 2.4 kb BamHI fragment from pEC2.2 (lacking codons for the first 76 aa of cellulase, including the 31 aa signal peptide) into the BglII site of pMEL.D.9.1 (ATCC 53360).
An additional codon (AAG, lysine) was present between the terminal codon (GGG, glycine) of the melibiase signal peptide and codon 77 (ATC, isoleucine) of cellulase.
ATCC 53361, 53362, and 53363 contain the same 6.7 kb BamHI fragment which comprises the operative melibiase:endoglucanase gene fusion.
