Restriction digests of the clone give the following sizes (kb): PstI--2.8, 1.4, 0.58; BamHI--2.9, 1.9; EcoRI--3.6, 1.2.
The vector contains the marker sup3-5, which suppresses ade6-704, and the tet repressor gene under control of the S. pombe adh promoter.
When integrated into a strain of genotype h- ade6-704 leu1-32 ura4-D18, the resulting fission yeast strain, FY191 (ATCC ) produces a constitutive tetracycline repressor protein.
