Diagnosis of ATRT was validated by immunohistochemistry that demonstrated negative expression of INI-1 in the tumor cells, but normal expression by endothelium on the same section.
This cell line has been continuously carried in culture for over 18 months, and in neurobasal medium grows as spheres of varying sizes.
The generation of this cell line was made possible with the support of Grayson's Fund and the Michael Hoefflin Foundation for Children's Cancer to Childrens Hospital Los Angeles, with the goal of making pediatric brain tumor lines available to the research community.
Cultures can be maintained by the addition of fresh medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension in ¼ volume of the conditioned medium and ¾ volume fresh medium at 2 X 105 to 3 X 105 viable cells/mL. Do not allow the cell concentration to exceed 2 X 106 cells/mL.
Subcultivation ratio: A subcultivation ratio of 1:4 to 1:6 is recommended.
