The insert contains the following sites separated by (kb) (approximate): EcoRI- 1.0- EcoRI- 0.1- SphI- 0.3- EcoRI- 1.0- XhoII- 1.6- EcoRI- 0.3- XbaI- 1.1- XhoII. Restriction digests of the clone give the following sizes (kb): SmaI/SalI--6.2, 4.2, 4.1; SalI--10.6, 4.1; EcoRV--14.4; XbaI--9.3, 5.2. The smallest subclone capable of complementing a cor1-1 mutation was a 3.0 kb XhoII fragment. This region contains a continuous 1.368 kb open reading frame encoding a protein of 456 amino acids with a molecular weight of 50 kDa. The probe, pG7/T1, has been deposited in Escherichia coli RR1 as ATCC 37665 and in Saccharomyces cerevisiae N5-7/U7 as ATCC 66316. |
