This conditional lethal mutant, originally designated plaque isolate no. 3679, was isolated from the IHD-W strain of Vaccinia by S. Dales. The IHD-W strain was derived from an IHD-J strain of Vaccinia.

Selected after mutagenesis with nitrosoguanidine. The non-permissive incubation temperature is 39.5 C. This mutant was assigned a Dales EM categories G and P, characterized by incomplete membranes with spicules, immature normal particles and mature particles with cores that are predominantly lucent at the center. The mutation was assigned by complementation analysis to vaccinia map location A24, the 132-kDa viral RNA polymerase subunit.

Effect on Host

CPE: enlargement, plaques with rounding and eventual sloughing

Cytopathic effects in cell culture include cell rounding and death, with eventual sloughing
